负载阿霉素的Ti基金属有机框架/壳聚糖支架的制备及其在骨再生和抗肿瘤中的应用

摘要:手术切除是骨肿瘤的主要治疗方式;然而,它造成局部骨缺损和易导致肿瘤复发。因此,骨肿瘤术后迫切需要兼具治疗与修复的生物材料。本研究提出合成具有超高比表面积的Ti-MOF,然后将负载阿霉素(DOX)的Ti-MOF,与壳聚糖(CS)溶液混合,冻干形成支架。研究Ti-MOF的促进成骨分化性能,Ti-MOF的载药量,DOX@Ti-MOF的抗肿瘤活性及其植入支架材料后支架结构变化。结果表明,Ti-MOF比表面积高达1116 cm3/g,并且具有很好的生物相容性以及促成骨分化能力;Ti-MOF的载药量为DOX/Ti-MOF:6.83 mg/g;DOX@Ti-MOF具有出色的抗肿瘤活性。扫描电镜结果显示:DOX@Ti-MOF的加入对支架结构无明显的影响。基于DOX@Ti-MOF良好的促进成骨分化能力以及抗肿瘤活性,使得DOX@Ti-MOF/CS可能是一种有前途的术后骨肿瘤清除及骨缺损修复平台。

Preparation of Ti-based metal organic framework/chitosan scaffold loaded with adriamycin and its application in bone regeneration and antitumor
Abstract:Surgical resection is the main treatment modality for bone tumors; however, it causes local bone defects and predisposes to tumor recurrence. Therefore, biomaterials that combine both treatment and repair are urgently needed after bone tumor surgery. In this study, we propose to synthesize Ti-MOF with ultra-high specific surface area, and then Ti-MOF loaded with adriamycin (DOX), mixed with chitosan solution and lyophilized to form scaffolds. The osteogenic differentiation-promoting properties of Ti-MOF, the drug loading of Ti-MOF, the anti-tumor activity of DOX@Ti-MOF and its structural changes of the scaffold after implantation into the scaffold material were investigated. The results showed that Ti-MOF had a high specific surface area of 1116 cm3/g and excellent biocompatibility and ability to promote bone differentiation; the drug loading of Ti-MOF was DOX/Ti-MOF: 6.83 mg/g; and DOX@Ti-MOF has excellent anti-tumor activity. SEM results showed that the addition of DOX@Ti-MOF had no significant effect on the scaffold structure. Based on the good osteogenic differentiation-promoting ability and anti-tumor activity of DOX@Ti-MOF, it makes DOX@Ti-MOF/CS may be a promising platform for postoperative bone tumor removal and bone defect repair.
